The Experimental Studies of the Influences of the Testis upon the Allergic Changes of the Joint-capsule


Abstract
There have been numerous invessfigations and reports concerning effects of endocrine organs on experimental allergic reactions of various organs. However, reported results have been variable in many respects and sometimes caused certain confusions. This investigation deals with an experimental study of allergic inflammation produced in joint-capsule and to study effects of testis and its hormone upon such inflam;nations.
MATEIRIALS AND METHODS
Healthy male albino rabbits weighing around 2kg were used. The animals were fed with an ordinary diet consisted of bean curd and supplemented with vegetable pieces. The antigen employed was fresh horse serum prepared by the laboratory of the National Bureau of Preventive Medicine. The testosterone utilized was Testoviron made by Schering AG of Berlin, West Germany.
The animals were divided into six groups as the following:
1. Normal control group.
2. Animals with orchidectomy.
3. Animals with orchidectomy and also receiving auto-transplantation of testis into the thigh muscles.
4. Animals with orchidectomy and receiving intramuscular injection of testosterone.
5. Animals with homo-transplantation of testis into the thigh muscles,
6. Animals receiving testosterone injection.
Each group of animals received two sets of experimentations. One set of experiment consisted of initial sensitization by horse serum given subcutaneously 1.0 cc per kg body weight every other day for four times. After two weeks from the completion of the initial sensitization, 0.1 cc per kg body weight of horse serum was injected into the joint capsule. Another set of experiment consisted of injection of horse serum into the joint capsule 0.1 cc per kg body weight.
After the completion of injections, animals were sacrificed at regular intervals of 3 hours, 6 hours, 12 hours, 24 hours, 48 hours, 72 hours, one week and 2 weeks respectively. Air embolism was exclusively used for the method of sacrifice. Proper amount of joint capsule wds resected and fixed in 10% neutral formalin and stained with hematoxylin & eosin and Van-Gieson stains.
RESULTS AND SUMMARY
1. Experimental allergic inflammation has been produced in the joint capsule of albino rabbits by using horse serum as antigen and an attempt has been made to investigate the effects of testis and testicular hormone upon such inflammations.
2. Careful histological analysis reveals the intensity of inflammatory reaction to be in the order of a). animals receiving testosterone injection, b). animals with homo-transplantation of testis, c). normal control group, d), animals with orchidectomy and receiving testosterone, e), animals with orchidectomy and receiving auto-transplantation of testis and f), animals with orchidectomy respectively.
3. From the results obtained by above experiments, it is the opinion of this investigator that the testis and testicular hormones definitely enhance experimental allergic inflammation of joint capsule.
